背景技術
[0002] キノロン系合成抗菌剤は、ノルフロキサシンの発見以来、抗菌活性や体内動態が 改善され、ほぼ全身の感染症に有効な化学療法剤に発展し、多くの化合物が臨床 の場に供されている。
[0003] しかし、近年、臨床の場ではキノロン系合成抗菌薬に対して低感受性菌が増カロしつ つある。例えば、グラム陽性菌において、 j8—ラタタム系抗生物質に非感受性の黄色 ブドウ球菌 (MRSA)や肺炎球菌 (PRSP)、そしてアミノ配糖体系抗菌薬に非感受性 の腸球菌 (VRE)の如くキノロン系合成抗菌薬以外の薬剤に耐性の菌であって、更 にキノロン系合成抗菌剤に低感受性の菌も増加している。従って、臨床の場では有 効性がさらに高い薬剤が望まれている。
また、非ステロイド性の抗炎症剤との服用による痙攣、光毒性等の副作用も明らかと なっており、より安全性の高 ヽキノロン系合成抗菌薬の開発が求められて!/、る。
[0004] キノロン系合成抗菌薬の抗菌活性、体内動態そして安全性には、キノロン骨格の 7 位及び 1位の置換基の構造が大きく関与することが知られて 、る。キノロン骨格の 7 位が 3—ァミノメチルピロリジ -ル基で置換されたキノロン誘導体は、グラム陰性菌及 びグラム陽性菌に対して強い抗菌活性を有することが知られている。例えば、 7— [3— (1 アミノメチル)ピロリジン 1 ィル]キノロンカルボン酸誘導体 (非特許文献 1参照) である。更に、当該アミノメチル基が更に置換されたキノロンカルボン酸誘導体として 、 7— [3— (1 アミノエチル)ピロリジン 1 ィル]キノロンカルボン酸誘導体 (非特許文 献 2参照)、 7—[3—(1 アミノー 1ーメチルェチル)ピロリジン 1 ィル]キノロンカルボン 酸誘導体 (非特許文献 3参照)、 7-[3- ( 1-アミノアルキル)ピロリジン- 1-ィル]キノ ロンカルボン酸誘導体 (非特許文献 4参照)等が知られて ヽる。
[0005] しかし、上記のキノロンカルボン酸誘導体の多くは選択毒性が低!、ために、細菌だ けでなく真核生物の細胞に対しても作用し (非特許文献 5参照)、医薬又は動物薬と しての使用は困難であり、実際にこれまで臨床に供されたものはない。
[0006] 一方、本願発明に関連するキノロン骨格の 7位が 3—(1 アミノシクロアルキル)ピロ リジ-ル基で置換されたキノロンカルボン酸誘導体 (A) (特許文献 1参照)及び (B) ( 特許文献 2参照)が知られている(尚、これらの化合物における置換基の定義は、そ れぞれ特許文献 1、 2において定義されたものであって、同じ記号であっても本願明 細書における置換基の定義とは無関係である)。

[0009] し力しながら、これらの出願において具体的に開示されたキノロンカルボン酸誘導 体は、キノロン骨格の 8位カ チル基もしくはメトキシ基で置換、あるいはメトキシ基が キノロン骨格の窒素原子を含んで環を形成している化合物のみである。また、上記の 化合物は、従来のキノロン誘導体に比較して強力な抗菌活性を示すが、急性毒性が 強ぐかつ遺伝毒性の指標である小核試験において総じて陽性を示す。

[0014] 従って、本発明は、グラム陽性菌及びグラム陰性菌に対して強力な抗菌活性を示し 、しかも高 、安全性を有するキノロン系抗菌薬及び感染症治療薬を提供することを目 的とする。
課題を解決するための手段
[0015] 本発明者らは、斯カる実情に鑑み、抗菌力に優れ、かつ安全性の高いキノロン系 化合物を得るべく鋭意研究した結果、下記式(1)で表わされる 8—シァノキノロンカル ボン酸誘導体、その塩又はそれらの水和物が、グラム陽性菌及びグラム陰性菌、特 に MRS A、 PRSP及び VREを含むグラム耐性腸球菌に代表される耐性菌に対して、 公知のキノロン系化合物と比較して強力な抗菌活性を示し、更に抗菌薬として高い 安全性を兼ね備えていることを見出し、本願発明を完成させた。

[0120] 表 1及び 2から明らかなように、本発明の化合物は、耐性菌を含めたグラム陽性菌及 びグラム陰性菌の何れに対しても幅広くかつ非常に強力な抗菌活性を示した。

化合物番号 1及び 2の化合物について、マウス骨髄小核試験を以下に記載の方法 にて実施した。
6週齢 Sic: ddY系雄性マウスを各群 5匹ずつ使用し、化合物番号 1及び 2の化合物 を 0. ImolZlNaOHZ生理食塩水で溶解、希釈した。コントロールとして 0. lmol ZlNaOHZ生理食塩水溶媒、陽性対照薬剤としてシクロホスフアミドを生理食塩水 で溶解、希釈した薬液を用いた。いずれもマイレックス GS 0. 22 mフィルターで 濾過滅菌した。各薬液を 10mLZkg、 0. 2mLZminの投与速度で単回静脈投与( 100、 150mgZkg)した。投与後 24時間に大腿骨力も骨髄細胞を採取、塗沫標本 を作成し、アクリルオレンジで染色した。蛍光顕微鏡で個体当たり 1000個の多染性 赤血球を観察し、小核を有する多染性赤血球の出現頻度及び赤血球 1000個中の 正染性赤血球と多染性赤血球の比を計数した。
その結果、特に本発明の化合物番号 1の化合物は、 150mgZkgの投与群におい て、コントロールと小核誘発率に有意差は見られず、判定結果は陰性であった。すな わち、本発明の化合物番号 1の化合物は、小核誘発作用が極めて弱ぐ安全性が高 いことが判明した。
